Apoptosis assay cell type - THP-1

A gross majority of classical apoptotic attributes can be quantitatively examined by flow cytometry, the preferred platform for rapid assessment of multiple cellular attributes at a single-cell level. However, sample preparation for such flow cytometry-based techniques could be challenging. Cell harvesting by trypsinization, mechanical or enzymatic cell disaggregation from tissues, extensive centrifugation steps, may all lead to preferential loss of apoptotic cells. To overcome this strictly follow manufacturers instruction of the detection kit.

Downstream tips
- When the absorbance of
samples too high, reduce agent-concentration and decrease incubation period.

- When the absorbance of
samples too low, increase the agent concentration and prolong the agent incubation interval
Upstream tips
- 1×10^6 cells were seeded and infected
Protocol tips
- Cells were double stained with Annexin-V-FITC and propidium iodide (PI) at different time points after infection
Upstream tips
- 2 × 10^5 cells were cultured
Protocol tips
- Annexin V‐FITC is treated for 30 min and then with propidium iodide (PI) for 5 min at room temperature,
