Annexin V-PE/7AAD staining
PC cells were grown at a density of 50% confluence in 100-mm culture dishes and were treated with varying concentrations of the drugs for 48 hours. Cells were stained using Annexin V-PE apoptosis detection kit (BD-Pharmingen) according to the manufacturer’s recommendations. Briefly, the cells were washed in PBS and resuspended in binding buffer (1×106 per ml). 5µl of AnnexinV-PE and 5µl of 7-AAD (7-aminoactinomycin-D) were added and incubated for 15 minutes at room temperature in the dark and analyzed by flow cytometry.
