Hello! Quick question for you guys. Can T4 DNA ligase join non-cohesive ends if my plasmid has overhang nucleotides on both ends?
Experiment: DNA isolation / purification Yeast - Cryptococcus neoformans
Your plasmid should not be able to be ligated if the nucleotides at its ends are non-cohesive. T4 DNA ligase can seal plasmids with cohesively ended substrates and blunt-ended substrates.
