DNA Ligase in duplex DNA is forming a phosphodiester bond between juxtaposed 5' phosphate and 3' hydroxyl termini. In order to perform this reaction, it needs ATP. If ATP is not present, the phosphodiester bond won't form and the two ends of DNA won't hold.
