Transfection is a powerful technique that enables the study of the function of genes and gene products in cells. Based on the nature of experiments, we may need a stable DNA transfection in cells for persistent gain-of-function or loss-of-function of the target gene. For stable transfection, integration of a DNA vector into the chromosome is crucial which requires selective screening and clonal isolation. By carefully selecting a viral delivery system and related reagents we can ensure safe and highly-efficient delivery of expression constructs for high-level constitutive or inducible expression in any mammalian cell type.

jetPEI-HUVEC/DNA mixture was added drop-wise onto the M199M+Glutamax™ I medium (GIBCO) containing 2% FBS, mixed gently, and incubated in a humidified 37°C incubator for 4 h. The growth medium was then replaced and the cells were incubated further for 24 h.
Downstream tips
To verify the transfection efficiency, HUVEC was transfected with Thy-1-pCMS-EGFP (enhanced green fluorescent protein)-C1, and then monitored using an inverted fluorescent microscope.

The second protocol uses the Targefect -HUVEC (Targefect F-2) reagent along with the Peptide enahncer and requires that cell be transfected in the absence fo serum.
