U251 cells (5×105/well) were seeded into 6-well plates containing antibiotic-free DMEM (HyClone; GE Healthcare Life Sciences, Logan, UT, USA) the day prior to transfection. The cells were transfected with 50 nmol/l CCL2 siRNA or the negative control using Lipofectamine™ 2000 (Invitrogen; Thermo Fisher Scientific, Inc., Waltham, MA, USA), according to the manufacturer's protocol. The sequence of the CCL2 siRNA was 5′-CTCGCGAGCTATAGAAGAA-3′.
