Plasmids and purification: The 12-LO/pcDNAI vector containing the full-length (2.0 kb) mouse leukocyte 12-LO cDNA under the control of the cytomegalovirus promoter was a gift from Dr. Colin Funk (University of Pennsylvania, Philadelphia, PA). The empty vector pcDNAI was purchased from Invitrogen (Carlsbad, CA) and used as mock transfection of 12-LO cDNA. Luciferase reporter gene construct pB2–1521 plasmid containing −1521 to +2 bp of the human CYP11B2 promoter sequence was cloned into the pGL3-basic luciferase reporter plasmid (Promega Corp., Madison, WI) in Dr. Rainey’s lab. The pRSV-LacZ plasmid expressing β-galactosidase as internal reference plasmid was purchased from Stratagene (La Jolla, CA). All plasmids for transfection were purified by EndoFree plasmid mega kit (QIAGEN Inc., Valencia, CA) following the manufacturer’s protocol.
