RNA extraction and sample handling: RNA extraction was performed using the RNeasy DSP FFPE kit on 4 to 5 μm thick FFPE sections with a tumor area between 100 and 500 mm2. Macrodissection was performed if necessary to attain a minimum of 80% tumor area. All steps outlined in the RNeasy DSP FFPE kit protocol were performed at room temperature (15 to 25 °C). Once extracted the RNA samples could either be tested immediately with the RT‐PCR kit or stored at −90 to −65 °C and a maximum of five freeze–thaw cycles were permissible.Full paper Login or join for free to view the full paper.

RNA isolation from tissue
How do I extract RNA from animal tissue without using liquid nitrogen? I tried the RNA extraction by using the TRIzol reagent and I homogenize the tissue using polytron homogenizer at room temperature for 30secs is this correct?

Problem in phase separation after using serum/plasma kit
I used a serum/plasma kit for my serum samples. After the phase separation the samples should have 3 phases: a colourless aqueous phase, a white interphase and a red organic phase. However, in some of my samples there was no aqueous phase unless I wait for an extended period of time. How can I circumvent this problem?
